This course is addressed to biochemists and chemists interested in biomolecules. It will present practical (through 21 hours of tutorials, experimental and computational approaches including some on the ESRF beamlines) and theoretical (through 19 hours of lectures) aspects of the classical approaches used to determine the structure of biomacromolecules by X-ray crystallography and Nuclear Magnetic Resonance. Students will be introduced in the different steps of the resolution of a structure of a biomolecule at the atomic scale, with an emphasis on proteins and DNA. The course will start on the evaluation of the sample quality and educate the audience towards decision-making in structural work. After an introduction to the physical principles of the two techniques, students will gain expertise in experimental structure determination and develop critical analysis of the obtained structural models.
